Genetic laboratory
The micropalaeontology group at MARUM operates a fully equipped laboratory for single-cell genetic analysis of marine microorganisms. Our primary target are planktonic and benthic foraminifera and their algal symbionts. The genetic analyses are carried out in two different laboratories dedicated to specific steps in the production of DNA sequence data. Our laboratories are certified to the S1 standard to allow DNA sequence amplification by cloning.
The work conducted in the pre-PCR Lab (Room 1680) involves DNA extraction from individual foraminifera and preparation of PCR mix to amplify the DNA. To avoid contamination of the single cells by foreign material, the DNA extraction is carried out under a clean bench and the preparation of the PCR mix under a UV hood.
The post-PCR Lab (Room 1770) is dedicated to work with high concentration of DNA, including PCR amplification of specific genes, screening of the PCR products and cloning. To this end, the laboratory is equipped with thermocyclers, electrophoresis gels, a UV-chamber for gel inspection equipped with a camera and a nannodrop for DNA quantification. PCR products are either sent directly for sequencing or cloned. All sequencing is carried out by external providers.
