The outline of the PhD is to analyze organic samples (i.e. dissolved and particulate organic compounds, microbial biomass, and endosymbiotic bacteria within organisms) taken from submarine hydrothermal systems which represent the deepest, hottest and harshest of Earth’s environments known to date. Valuable and challenging information on ecosystems that function independently of photosynthesis and where organisms are living close to the limit of life will shed light into the interaction between geochemical and biological processes in the deep ocean and seafloor sediments. A wider knowledge of Earth’s submarine biosphere including high productive hydrothermal sites is of potential importance. Furthermore, investigations of hydrothermal vent community structures might contribute to our knowledge about how life may have originated on Earth.


Master-thesis

Lipid Biomarkers of Thiotrophic and Methanotrophic Symbionts in different Bathymodiolus mussel species

In my Master thesis I investigated the ecology and biogeochemistry of endosymbiotic bacteria (i.e., thiotrophic and methanotrophic bacteria) in seep- and vent- mussels. The symbiont-containing gill and symbiont-free foot tissue of B. childressi and B. brooksi from the Gulf of Mexico (GoM) and B. cf. thermophilus from the Pacific Antarctic Ridge (PAR) were analyzed with different lipid analysis methodologies. Intact polar lipids (IPLs) mainly provided information on general compositional pattern; bacteriohopanepolyols (BHPs) indicate the input from bacterial sources and organism-specific biomarker (apolar lipids: FAs and sterol) and their isotopic composition demonstrated the symbiotic relationship of sulfide- and methane-oxidizing bacteria within the host organism.
The IPL- and BHP- biomarker analysis was conducted with HPLC/MSn whereas the apolar biomarkers were analyzed with GC/MS and GC-irMS.
